Trump was arrested on criminal charges related to hush money payments made to adult film actress Stormy Daniels in 2016.
Rock spoke out against the move during his appearance at the Mark Twain Prize on Sunday, where he was helping to honor Adam Sandler.
He addressed the lawmakers present in the audience, which included Nancy Pelosi, asking if they were really going to arrest Trump, and warning that doing so would only make him more popular.
Rock also joked about the charges that Trump faces, saying that paying off someone to keep quiet about an affair was actually quite romantic.
Trump has pleaded not guilty to the 34 criminal charges against him, and was taken into police custody before his upcoming arraignment.
